Impact of pollutants on aneuploidy in the Pacific oyster, Crassostrea gigas ArchiMer

Aneuploidy is the alteration of the normal diploid chromosome number. In the Pacifie oyster, Crassastrea gigas, hypodiploid cells have regularly been reported as have a negative correlation between this phenomenon and growth and evidence for a genetic basis. We previously demonstrated a positive relationship between a pollutant, atrazine, and aneuploidy in Crassastrea gigas adults and juveniles. A toxic metal, cadmium (Cd), can accumulate in human organs. Placenta is usually used as indicator organ for Cd exposure. Therefore, we aim to investigate the different of placental morphology between the low- and high-Cd accumulated placentas. The samples were collected from 14 pregnant women who resided in low-Cd contaminated (L-Cd group) and high-Cd contaminated (H-Cd group) areas. The concentrations of Cd in blood (B-Cd), urine (U-Cd) and placentas (P-Cd) were measured by ICP-MS and AAS. Influence des facteurs environmentaux sur l'aneuploïdie chez l'huître creuse, Crassostrea gigas ArchiMer

Différents travaux menés depuis 1984 chez l'huître creuse en particulier, montrent qu'i existe, dans les branchies (Thiriot-Quiévreux C., 1982), une certaine proportion de cellules possédant un nombre de chromosomes inférieur au nombre habituel 2N=20. Ce phénomène est l'aneuploïdie. Cette anomalie cytogénétique est mise en évidence par un comptage de chromosomes d'une suspension cellulaire obtenue à partir de branchies. Il est apparu que le taux d'aneuploïdie affectait négativement les taux de croissance (Leitao & al. 2001). Il existe deux types d'origines possibles à l'aneuploïdie. The impact of supplemental UV-B (sUV-B) radiation and heavy metals: Cd (68 µmol.kg-1 soil) and Ni (68 µmol.kg-1 soil) singly and in combination was studied on photosynthetic and non-photosynthetic pigments, metabolites, enzyme activities, biomass accumulation and the net primary productivity of spinach (Spinacia oleracea L. var. All Green), a common leafy vegetable plant. Both the stresses individually and in combination resulted in the reduction of photosynthetic pigments, ascorbic acid, and catalase activity, whereas a reverse trend was observed for anthocyanin, flavonoids and proline content, lipid peroxidation and peroxidase activity. Interspecific comparison of Cd bloaccumulation in European Pectinidae (Chlamys varia and Pecten maximus) ArchiMer

The uptake and loss kinetics of Cd were determined in two species of scallops from the European coasts, the variegated scallop Chlamys varia and the king scallop Pecten maximus, following exposures via seawater, phytoplankton and sediment using highly sensitive radiotracer techniques (Cd-109). Results indicate that, for seawater and dietary pathways, C. varia displays higher bioaccumulation capacities in terms of uptake rate from water and fraction absorbed from ingested food (assimilation efficiency) than Pecten maximus. Regarding sediment exposure, P. maximus displayed low steady-state Cd transfer factor (TFSS < 1); however, once incorporated, a very large part of Cd transferred from sediment (92%) was strongly retained within R maximus tissues....

La contamination de la Gironde par le cadmium : origine, extension, importance ArchiMer

In 1979, the French monitoring program RNO (Réseau National d'Observation de la qualité du milieu Marin) pointed out a dramatic cadmium contamination in the oysters from the Gironde estuary (up to 100 ug -1, dry weight). The wild oyster-beds upstream the estuary were shown to be the most contaminated. This result tends to point to the river as the source of the cadmium contamination. At the same rime, uncontaminated oysters from Arcachon were transplanted in the Gironde estuary (La Fosse, Dau). This report synthesizes the knowledge on the biogeochemical cycle, bioconcentration processes and ecotoxicity of cadmium in the marine environment. Man induced changes on the cadmium cycle, especially along the French coasts are reviewed. Concentration levels, mechanisms and fluxes between geochemical reservoirs are assessed. Emphasis is given on sublethal effects and toxicity mechanisms. Quality standards used in European Community countries are listed. Conclusions and recommendations for environmental management and research are given.

Lead (Pb2+) and cadmium (Cd2+) inhibit the dipsogenic action of central beta-adrenergic stimulation by isoproterenol BJMBR
We have previously demonstrated that acute third ventricle injections of both Pb2+ and Cd2+ impair the dipsogenic response elicited by three different situations: dehydration and central cholinergic or angiotensinergic stimulation. ß-Adrenergic activation is part of the multifactorial integrated systems operating in drinking behavior control in the central nervous system. Les études sanitaires et la pêche à pied professionnelle : l'exemple de Bonne Anse en Charente-Maritime ArchiMer

Tout coquillage mis en vente est issu d’une zone de production (gisement naturel ou concession) classée et surveillée au niveau sanitaire. Quatre classements sont définis de A à D par ordre décroissant de salubrité, du classement A (bonne qualité) autorisant la commercialisation directe des coquillages, au classement D (très mauvaise qualité). Pour définir ce classement, une étude sanitaire est préalablement conduite par l’Ifremer sur demande de l’Administration. L’étude sanitaire réalisée à Bonne Anse en Charente-Maritime, a abouti en 2009 à un arrêté préfectoral classant en B le gisement de palourdes. Aujourd’hui, la qualité sanitaire de la zone est surveillée régulièrement par l’Ifremer, et environ 25 pêcheurs à pied professionnels exploitent le gisement.

Dans l’objectif de déterminer l’état de la contamination organique et inorganique des sédiments superficiels de la frange côtière du golfe de Gabès, nous avons évalué les concentrations en hydrocarbures aromatiques totaux, en cadmium (Cd), en plomb (Pb) et en mercure (Hg). L’échantillonnage a été effectué à bord d’une barque motorisée, à l’aide d’une benne Van Veen au niveau de 25 stations. Long-term records of cadmium and silver contamination in sediments and oysters from the Gironde fluvial-estuarine continuum - Evidence of changing silver sources ArchiMer

Metal bioaccumulation and metallothionein concentrations in larvae of Crassostrea gigas ArchiMer

Larval stages of bivalve molluscs are highly sensitive to pollutants. Oysters from a hatchery from Normandy (English Channel) were induced to spawn, and fertilized eggs were exposed to copper or cadmium for 24 h. Metal accumulation (from 0.125 to 5 mu g Cu L-1 and from 25 to 200 mu g Cd L-1) and MT concentrations were measured in larvae. Metallothionein: An Overview on its Metal Homeostatic Regulation in Mammals International Journal of Morphology

Metallothionein (MT) is a ubiquitous protein with a low molecular weight of 6-7 kDa weight and it was first identified in the kidney cortex of equines as a cadmium (Cd)-binding protein responsible for the natural accumulation of Cd in the tissue. The mammalian MT contains 61 to 68 amino acid residues, in which 18 to 23 cysteine residues are present. The expression of MT starts by binding of metal transcription factor-1 (MTF-1) to the regulative region of MT gene called metal responsive elements (MREs). The induction of MT through the MREs region can be initiated by several metal ions such as zinc (Zn), copper (Cu) and Cd. Metallothionein mRNA induction is correlated with the decrease of DNA strand breaks in cadmium exposed zebra mussels ArchiMer

We have previously shown that cadmium (Cd) and Benzo[a]pyrene (BaP) induced early DNA damages in zebra mussels, and that the level of DNA strand breaks (SB) returned to a basal level after 3 days of exposure to Cd. The aim of the present study was to go further in the mechanisms of Cd and BaP detoxification. For that purpose, expression of genes encoding for metallothionein (MT), Aryl Hydrocarbon Receptor (AHR), P-gP, catalase, glutathione S-transferase and Heat shock protein 70 (HSP70) proteins have been measured using RT-qPCR. Data reported here show that Cd is a strong inducer of MT and HSP70 genes, and that BaP is a strong inducer of P-gP and AHR genes. Mise au point d'un test microcrustacé marin ArchiMer

Because of the low number of currently available toxicity tests for marine environment, the authors describe an experimental procedure using a marine copepod : Tigriopus brevicornis. This method can be used to quantify the 50 % inhibition of larval production or growth. The variability in controls larval production reaches about 9 %. The toxicity of two contaminants : tributyltin (TBTF) and cadmium chloride (CdCl2) was determined with this method and the 50 % decreases in larval production are respectively 1,4 µg/l (TBTF) and 61 µg/l (Cd++).

New insights from age determination on toxic element accumulation in striped and bottlenose dolphins from Atlantic and Mediterranean waters ArchiMer

This study aimed at comparing toxic element (Hg, Cd) bioaccumulation in relation to age for bottlenose (Tursiops truncatus) and striped dolphins (Stenella coeruleoalba) from Mediterranean and Atlantic waters. Metal concentrations were also measured in selected prey to infer metal exposure through the diet. As expected, Mediterranean prey exhibited the highest Hg levels, probably as a consequence of the Hg enrichment of the Mediterranean Sea. Comparing the predators from each area and taking age into account, Mediterranean bottlenose dolphins displayed higher Hg levels than Atlantic dolphins (p = 0.032), whereas Mediterranean striped dolphins did not (p = 0.691).
